Incidence of group A rotavirus in urban and rural areas of the city of Londrina-Brazil, from 1995 to 1997 BABT
Meneghetti,Ângelo Cesar; Bolognini,Andrea Maria; Lauretti,Flávio; Linhares,Rosa Elisa Carvalho; Santos,Norma; Nozawa,Carlos Mitihiko.
Rotaviruses are common pathogens and the causal agents of acute diarrhea among children and young animals. The involvement of rotavirus in human diarrheal disease among population of urban and rural areas of the city of Londrina, Parana was evaluated. Nine hundred and five fecal specimens from persons with diarrhea were studied, being 686 and 219 from urban and rural areas, respectively. Thirty-eight samples (4,2%) were positive for rotavirus by polyacrylamide gel electrophoresis of viral RNA and latex agglutination test of which 36 were from urban and two from rural areas. Out of the positive specimens, 17 strains were further characterized by RT-PCR typing assay, resulting in 16 strains of G1 genotype while one sample was found to be a mixture of G1 and...

Molecular epidemiology of the human group A rotavirus in the Paraná State, Brazil BABT
Santos,Jucélia Stadinicki dos; Alfieri,Alice Fernandes; Leite,José Paulo Gagliardi; Skraba,Irene; Alfieri,Amauri Alcindo.
From January/2000 to December/2003, 550 diarrheic fecal samples from the children and adults were collected in several geographical regions of Paraná State, Brazil. The enzyme immunoassay showed 120 (21.8%) samples positive for the group A rotaviruses. One hundred and fourteen samples were genotyped by multiplex-nested-PCR assay. The highest frequency (77.5%) of the positive samples (n=93) was observed in the children under 5 years old. Rotavirus diarrhea was more frequent in the cold and dry seasons of the four evaluated years. The most frequent genotypes were: G1 (50.9%), G4 (9.6%), G9 (7.0%), G2 (1.7%), G3 (0.9%), P[ 8] (71.9%), and P[ 4] (3.5%). The P[ 8] G1 (46.5%) and P[ 8] G4 (9.6%) were the main combinations found to P and G genotypes. The mixed...

Molecular weight estimation of esterase isoenzymes in closely related Drosophila Species (Diptera: Drosophilidae) in non-denaturing polyacrylamide gel electrophoresis BABT
Mateus,Rogério Pincela; Cabral,Hamilton; Bonilla-Rodriguez,Gustavo Orlando; Ceron,Carlos Roberto.
A method that allows the measure of molecular weight of two well-known and closely related esterases from Drosophila mojavensis and its sibling species, D. arizonae, is here described, using native polyacrylamide gel electrophoresis at several concentrations, applying Fergunson´s principles. These enzymes, namely EST-4 and EST-5, presented molecular weight values between 81 and 91 kDa. In spite of their distinct expression pattern through the insect's life cycle, they showed properties of isoenzymes codified by distinct structural genes, supporting the hypothesis of a rather recent gene duplication event that generated both in D. mojavensis and D. arizonae, as well as in other species of repleta group. The method is simple and adequate to be applied to...

Padrões eletroforéticos de hordeínas e isoenzimas para identificação de cultivares de cevada Rev. bras. sementes
Binneck,Eliseu; Nedel,Jorge Luiz; Dellagostin,Odir A; Barros,A.C.S.A; Peske,Silmar Teichert.

Segmented double-stranded genomic RNA viruses in fecal samples from broiler chicken BJM
Tamehiro,Claudia Yurika; Alfieri,Alice Fernandes; Médici,Cristina; Alfieri,Amauri Alcindo.
Segmented double-stranded RNA (dsRNA) viruses were identified by polyacrylamide gel electrophoresis (PAGE) technique in fecal samples from broiler chicken. A total of 378 fecal samples from 1-7 weeks old chickens were analyzed. dsRNA with migration profile characteristic of avian rotavirus (AvRV), reovirus (ARV) or picobirnavirus (PBV) was identified in 32 (8.5%), 7 (1.8%) and 13 (3.4%) samples, respectively. AvRV and ARV occurred more frequently in chickens up to 1 month old and were related with enteritis signs. Considering only fecal samples of chickens with diarrhea, the AvRV was detected in 37.8% (14/37) and the ARV in 13.5% (5/37) of analyzed samples. AvRV was identified in only 1.5% (4/274) and ARV was not detected in normal feces collected from...

The occurrence of Orthoreovirus, Rotavirus and chicken anemia virus in chickens of the poultry industry in Minas Gerais, Brazil Arq. Bras. Med. Vet. Zootec.
Rios,R.L.; Marin,S.Y.; Gomes,A.M.; Resende,J.S.; Bonfim,M.R.Q.; Gomes,A.D.; Resende,F.M.S.; Horta,R.S.; Resende,M.; Martins,N.R.S..
Fifty-four fecal samples taken from broiler chickens from 1 to 45 days of age, and of pullets from 10 to 13 weeks of age, original from eight different poultry regions in the state of Minas Gerais, Brazil, were collected from March 2008 to January 2010 for avian Orthoreovirus (ARV) and avian Rotavirus (AvRV) analyses. For the assay of ARV, RNA was immediately extracted (Trizolâ) and transcribed into cDNA for assaying in a nested-PCR with ARV-specific primers. For AvRV, polyacrylamide gel electrophoresis (PAGE) was performed with RNA extracts obtained by phenol-chloroform extraction. CAV was additionally investigated through a nested-PCR of thymus and spleen. Results found 5.55% positive for ARV and 9.25% for AvRV. Also, CAV and ARV genomes were detected in...
